When measuring pressure or differential pressure, and depending on how the three (two, five) valve assemblies are installed, is it still necessary to add a stop valve? Are there any guidelines I can refer to?

A primary valve near one end of the equipment pipeline is still needed. It’s not necessary on the side closest to the transmitter; after all, the valve assembly is designed to replace the function of several valves.
